You have to select London in advance and save your file as default selection London. So when ever you open your file it will show London selection bydefaultm or you can make it as bookmark.

I want to create something that will force me on opening the Qlikview document to make a selection, based on location

so I have

north east

Midlands

London

Want to have to choose LONDON at start and then have all my charts linked to this selection so I guess using variables maybe but not really sure how to go about it??

Yes.because when you will open the file from access point .It will show you 1st the default save and after that you can change your selection midland to see the charts. In access point before open the qvw there is a option to remove LAST DOCUMENT STATE.After remove last document u can always strts with London but if u changes the selection in your development environment t and save it then it would show the access point according to your last qvw saved in development environment.
