Why does the removal of an equal sign in a variable declaration alters my result?

Why the result of a substraction changes between two variables (ECO-ECOLY)/ECOLY (Image 1) changes when I remove an equal sign (Image 2) in one of the variables (ECOLY) (Image 3)?

When a variable contains an equal sign in front of the expression, it will be evaluated globally with every selection.
