I am getting below error message when trying to open Nprinting Designer. Does anyone ever faced this kind of error and need help to resolve this issue.

You 'might' be encountering a reverse proxy configuration.
You can follow these steps if a reverse proxy is present.
Check with your IT team to confirm.
Also:
1. Ensure you have an NPrinting account in the domain or at the very least NPrinting security credentials. . Your NP admin will help you distinguish and which to use.
2. You should be part of the 'user' and 'developer' NPrinting security roles. Check with your NP admin
3. Ensure you are on the network where NPrinting is installed. (use a VPN if needed but you must be on the network)
4. If you are using a virtualized desktop ie: citrix or similar, this is not supported. Please contact your network admin. They will need to determine a workaround for you.
5. Ensure you have the NPrinting Designer and correct version installed directly on your computer. The version of NP designer must be the same version as the NP server.
You may need to check with your IT team if you are working remotely or if you have citrix desktop to arrange a custom solution for you to enable you to work with the NP designer.

We have not made any change on the server also we have tried & checked all the points mentioned above. Also we have not enabled any reverse proxy here as well. Could you please suggest why this happens ??

If this is affecting only your windows desktop, then I suggest you uninstall both MS Office and NPrinting, reboot and reinstall MS Office first, then NPrinting.
MS Office must be installed locally as well as NPrinting. Citrix desktops and Pure Cloud version of MS Offices are not supported.
If this is affecting all NP developers at your organization, then I suggest that you start a support case.
But is sounds like a localized problem on your desktop. (So also suggest you ask your IT department to run some network tests between your desktop and the NP Server that you are trying to connect to. (please remember that if you are external -outside the network connecting to NP server on the internal network ie: working remotely, your IT team will need to troubleshoot why it is not working for you).
All of this works very nicely if you are connecting to a valid internal NPrinting server from your desktop that are both on the same internal network and in the same domain.
