is the time that is stored contain seconds?  If seconds are not part of the field you will not be able to get accurate seconds (I do not believe)

=Interval(Time#('10:43:36 AM', 'hh:mm:ss TT') - Time#('10:22:00 AM', 'hh:mm:ss TT'), 's')

to convert sec's as well as minuts and hours and days also by using interval function
