I am not able to save changes what i have done in previously created QVW file.

OK, so you have a workaround for a File->Save that doesn't work.
Replace the original file with the copy only if you are absolutely sure that the copy contains everything from the original file (and your modifications).
I don't know why File->Save refuses to do its job. Are you sure no error messages are shown?

May be file is read only or due to any security check this with the System Admin people. And check you make changes to the script part or the application part.

Currently I am opening origional QVW file in production there is save option is not disabled and there is no section access is implemented.

When you use the "Open in Server" option then the QlikView Desktop works like a QV client which will allow you to open the file from the Server. (from Server's memory) the Save, Reload, Table Viewer, Edit Script options are disabled.
What happens if you select File->Save? Does QlikView Desktop throw an error? If you try File->Save As..., are you able to create a copy with a different name?

yes by doing save as it is creating other copy of qvw file
